So I planned to do the experiment that got derailed yesterday today. I set up cultures yesterday using new media and trying to be very careful to keep everything sterile. Everything seemed in order, and I expected to come in today to happy cultures. This morning I walked into the warm room to get my cultures. The first culture I pulled out looked perfect. No contamination. Lovely yeasty culture smell. Beautiful. I pulled out the second culture and what's this? There's a pipette tip in my culture. A pipette tip that I had not put in there. AHHHHH!!!! Some sleep deprived or just generally sloppy person added stuff to my culture instead of to their own culture!

O...k... no experiment today either. I did have a decent amount of cloning to do instead but not enough to keep me actively busy the entire day (as in, the full day more or less is required to do the stuff, but there were breaks for incubations). As it was once again a beautiful day, I decided that the best way to remedy the fact that I could not do my experiment was to go for a quick run with Presto. So I did. It was a perfect, gorgeous day for a run, and I am glad I got to get out in the sun. There were also people out by the river with this fancy camera set up looking all official and stuff and they took a bunch of pictures of Presto in his technicolored dreamcoat :-).
